上皮穩(wěn)態(tài)失衡與腫瘤
1.?上皮極性/穩(wěn)態(tài)調(diào)控與腫瘤
聚焦上皮極性基因的功能機(jī)制,系統(tǒng)解析以下四方面的細(xì)胞生物學(xué)問題:(1)極性信號(hào)在上皮穩(wěn)態(tài)維持中的核心作用;(2)極性信號(hào)與亞細(xì)胞結(jié)構(gòu)功能調(diào)控(例如線粒體和細(xì)胞骨架)的關(guān)系以及在上皮細(xì)胞能量代謝中的作用;(3)具有雙定位功能的極性信號(hào)行使入核調(diào)控功能的重要意義;(4)探究基質(zhì)剛度、組織壓力等力學(xué)微環(huán)境破壞上皮細(xì)胞功能、并促進(jìn)腫瘤進(jìn)展的力學(xué)生物學(xué)機(jī)制。通過以上四方面的研究,解析極性蛋白丟失協(xié)同致癌突變驅(qū)動(dòng)腫瘤發(fā)生的代謝重構(gòu)以及腫瘤微環(huán)境調(diào)控的機(jī)制。
2.?硒穩(wěn)態(tài)失衡與腫瘤發(fā)生
深入研究微量元素硒及硒蛋白家族因子在腫瘤氧化代謝壓力下的多層次調(diào)控規(guī)律,通過線粒體蛋白調(diào)控細(xì)胞死亡、炎癥以及與免疫代謝中全新機(jī)制探討,試圖解析腫瘤代謝重編程以及表觀遺傳調(diào)控規(guī)律,以明確其在腫瘤惡性轉(zhuǎn)化中通過對(duì)腫瘤細(xì)胞衰老等方式實(shí)現(xiàn)改善腫瘤免疫微環(huán)境的免疫抑制作用,將為硒抗腫瘤免疫干預(yù)提供新的臨床治療干預(yù)方案和新靶點(diǎn)。
3.?上皮穩(wěn)態(tài)與肝胰代謝性疾病
以肝胰上皮屏障為核心,研究上皮屏障紊亂與糖脂代謝微環(huán)境的相互調(diào)控機(jī)制,解析慢性代謝疾病中的免疫 - 代謝串?dāng)_規(guī)律,揭示肝屏障功能異常介導(dǎo)的以下病理生理過程:脂肪干細(xì)胞趨化分化等功能失衡、代謝紊亂與慢性免疫代謝微環(huán)境的關(guān)系及其炎癥的分子基礎(chǔ),從而發(fā)掘慢性肝病及代謝綜合征防治的新靶標(biāo)與新策略。
